I. Molecular Properties of NDTP:
NDTP is a mildly irritant, highly efficient condensation reagent operating under gentle reaction conditions. It catalyse the formation of amide and ester bonds within one minute, significantly enhancing reaction efficiency. Furthermore, NDTP is recyclable: reaction by-products can be recovered via simple filtration and reconverted into NDTP through a two-step process for reuse. This substantially reduces operational costs and aligns with green chemistry principles.

II. Mechanism of Action of NDTP:
The mechanism of action of NDTP is as follows: Under the action of a base, carboxylic acids react with NDTP to generate two reactive intermediates: activated carboxylic acid esters and acyl thiocyanates. These intermediates further react with amines or alcohols to efficiently generate the corresponding amide and ester compounds.

III. Functions of NDTP:
1. NDTP promotes amide synthesis
(1) Efficient and convenient dipeptide synthesis
NDTP can be used for dipeptide coupling reactions of various types of protected amino acids and amino acid methyl ester hydrochloride. It is suitable for nonpolar amino acids, polar amino acids and basic amino acids. Under mild conditions, the corresponding dipeptides are obtained in excellent yield within 1 minute without racemization and with good stereoselectivity.

(2) Easy construction of challenging amides
For substrates such as sterically hindered amines and aromatic amines that are difficult to convert efficiently by traditional methods, NDTP also performs well. It can achieve efficient amidation of various carboxylic acids and complex amines, with yields generally at a medium to high level.

(3) Fmoc-solid phase peptide synthesis
NDTP can also be applied to Fmoc-solid phase synthesis. On Rink Amide MBHA resin, the coupling of amino acids can be completed within 3 minutes, and this has been verified in 5-12 active peptides, fully demonstrating its high efficiency and compatibility in solid phase synthesis.

2. NDTP promotes ester synthesis
(1) Lower alcohols as solvents
In esterification reactions involving lower alcohols, they can be used directly as solvents. With the participation of NDTP, they can react with various carboxylic acids (such as nitrogen-containing heterocycles, dicarboxylic acids, unsaturated carboxylic acids, and various drug molecules) to form ester bonds. The yields are generally high, but the yields of electron-rich aromatic acids (such as methylbenzoic acid) are low.

(2) Acetonitrile as solvents
In esterification reactions involving higher alcohols, acetonitrile is used as a solvent. With the participation of NDTP, it reacts with carboxylic acids to form ester bonds. Its application range is wider and it is suitable for the efficient synthesis of primary alcohols, secondary alcohols, phenols, and lactones. Various drug molecules containing carboxyl or hydroxyl groups can also be esterified in good yields.
